Aim
  • The aim of the course is for participants to:
    1) Understand basic principles in flow cytometry
    2) Be able to perform flow cytometry experiments to determine various cell characteristics
    3) Gain knowledge of central aspects that are required for designing basic flow cytometrical experiments and selecting controls
    4) Be able to analyse flow cytometrical data
Contents
  • Flow cytometry is widely used for analysis of immune cell characteristics as well as for sorting of pure cell populations.
    This is a basic course in flow cytometry, which covers various applications of the technique, including analysis of extracellular and intracellular proteins, cell viability, cell proliferation, detection of proteins in solution and cell sorting. We will discuss important factors including compensation, selection and combination of fluorochromes and selection of appropriate control samples. The course also comprise a seminar to give basic understanding of how a flow cytometer instrument works.

    Practical parts will be performed on human immune cells in our cell lab, and samples will be analysed on BD FACS Canto and BD FACS Aria II instruments.

  • be able to describe basic principles in flow cytometry. Also, participants shall have general knowledge of various cell characteristics that can be assessed by flow cytometry.

  • be able to apply protocols for flow cytometry experiments to determine various cell characteristics.
    be able to analyse experimental results with software for flow cytometry data.
    be able to compare different protocols for flow cytometry and explain the different outcomes.
    be able to critically review flow cytometrical data in scientific papers.

  • The course is given during the autumn semester, within the Postgraduate Courses in the Life Sciences (http://www.cmps.lu.se/life_sciences) if at least 6 students are enrolled. Basic knowledge within the field of cell and molecular biology is expected. Some preparation before the course is expected and information on this will be provided via email before the course starts (e.g. installation of analysis program and reading).
